While driving through the desert, Grace Andrews (Sophia Bush) and Jim Halsey (Zachary Knighton), 2 college students, pick up a mysterious hitcher named John Ryder (Sean Bean), not knowing about his murderous plans.

- Rutger Hauer (who played the original hitcher) was originally planed to have a cameo in the beginning of the movie.
- At first the movie was planned to be released on April 16, but due to the early production it moved to February 18, and then to January 19.
- According to test screenings there are two endings. One where Grace walks to the nearest gas station and the other one where she gets killed in the last few seconds.
- Sophia Bush is the only survivor.
- The character Jim Halsey in the original version had the leading role but the character now has just a supporting role and gets killed in a car.
- Despite being a remake of The Hitcher, the plot of this film bears much more resemblence to it's direct to video sequel The Hitcher 2.
- Zachary Knighton beat out other actors to the role of Jim Halsey including Chris Pine, Jared Padalecki, and Justin Timberlake.
- Mary Elizabeth Winstead was offered the leading role but turned it down due to scheduling conflicts and commitments to Grindhouse. She also didn't want to become a typecast for a scream queen and wanted to take a break from horror movies.
- Sophia Bush barely beat out Hilarie Burton, Rachael Leigh Cook, and Elisha Cuthbert to the leading role.
- Kate Bosworth was offered the leading role but was forced to turn it down due to commitments to promoting Superman Returns which was very disappointing to her.
- Wes Craven was lined up to direct but pulled out.
- During early development, Kirsten Dunst was set to play Grace.
- Viggo Mortensen was offered the role of John Ryder but declined because he didn't want to be typecast as a horror icon.
