New York State Route 404 is an east-west running route following the former path of U.S. Route 104 in Monroe County. NY 404's eastern terminus is at NY 104 in Webster and its western terminus is at NY 590 in Rochester.

NY 404 begins at NY 590 as Empire Boulevard. Irondequoit Bay is northeast of this intersection, and Empire Blvd. serves as the northernmost non-bridge route from one side of the bay to the other (that is, between the inner-ring suburbs of Irondequoit and Brighton and the outer-ring suburbs of Webster and Penfield). Empire Blvd. heads east along the south shore of the bay for 1.5 miles, then turns northeast along a major commercial strip, another two miles, to the intersection with Ridge Road in West Webster. From there, NY 404 continues east as Ridge Road. This name remains for another three miles up to the village of Webster, where NY 404 becomes West and East Main St. The difference in direction refers to NY 404's position relative to NY 250 inside the village. East of Webster, Ridge Road returns and follows NY 404 east to the Monroe-Wayne County Line where, 1/2 mile before the county line, NY 404 turns north onto an unnamed road that sharply turns east, then north, merging into County Line Road and reaching its eastern terminus with NY 104.

The current alignment of NY 404 was assigned in 1972, replacing US 104 (now NY 104), which had been realigned to a newly constructed expressway to the north. Previously, NY 404 was used between Sleepy Hollow and Ossining on what is now NY 9A.
