New York State Route 376 is a New York state highway located entirely within Dutchess County. The route travels from Poughkeepsie, where it meets U.S. Route 44 and New York State Route 55, passing south through Red Oaks Mill and Hopewell Junction, ending in East Fishkill where it meets New York State Route 52.

NY-376 begins at NY-52 in Hopewell Junction and heads north to NY-82 where a short 30-foot duplex is formed. The road then heads east, then leaves 82 to go north, which are two of six intersections on 376 requiring a turn for through traffic. 376 then heads west at CR-29, then back north at CR-104. The route also meets NY-113 and CR-77.
Its exact route has changed a few times over the years, but it currently begins on Raymond Avenue in the town of Poughkeepsie (just outside the City of Poughkeepsie), then joins New Hackensack Road in a southeasterly direction just as it leaves the city (where its continuation in the other direction is called Hooker Avenue or Reference Route 983T). It then follows New Hackensack Road past the Dutchess County Airport but branches off to a different road soon afterward. It then winds through southern Dutchess County with several turns and forks until its end at East Fishkill.
