New York State Route 350 is a north-south running highway located entirely in Wayne County. The route, which acts as the eastern terminus for a handful of state routes, is one of the major north-south arterials in Wayne County, linking NY 104 to NY 31. The southern terminus of NY 350 is at NY 31 in the village of Macedon. Its northern endpoint is at NY 104 in Ontario Center.

Just east of the village of Macedon, NY 350 begins at an intersection with NY 31 near the former Mobil Chemical and Tyco plant, the largest business in the town. This intersection is also the eastern terminus of NY 31F, which duplexes with NY 350 north from this intersection, forming a technically unnecessary concurrency that lasts until Macedon Center, where NY 31F turns west toward Rochester while NY 350 turns to the east, then to the north.
North of Macedon Center, NY 350 receives its first and only name, Ontario Center Road. Farther north, in Walworth, NY 350 is the eastern terminus for two routes. First, at a four-way intersection with Walworth-Penfield Road, NY 441 comes to an end. Next, 1 3/4 miles to the north, NY 286 ends at a three-way intersection.
Between NY 286 and NY 104, NY 350 does not intersect any state routes. Instead, it crosses many local roads, swerving to the northeast, then to the northwest before meeting NY 104 in Ontario Center.
