Former Route 55 was a short highway in southeastern Missouri. Its northern terminus was at Route 9 (now U.S. Route 61) in Benton. Its southern terminus was at Wolf Island. It is now Route 77 from Benton to Charleston, and Routes 105, 75, 80, and Missouri Supplemental Route AA.

A branch highway, Missouri 55A connected the road to East Prairie and is now part of Missouri 105.
