Former Route 54 was one of the original 1922 state highways. Its eastern terminus was at Route 9 (now U.S. Route 61) in New London. Its western terminus was at Route 15 in Paris. The road was decommissioned to avoid confusion with U.S. Route 54 which runs nearby. The eastern half of the highway became part of Route 19 which was extended and the western half is now part of Route 154 which was designated after US 54.
A branch route, Missouri 54A, went south from Perry to the Ralls/Audrain County line. It became part of Missouri 19.
