Former Route 45 was one of the original 1922 state highways. It later became part of Route 19 when that highway was extended further north. Its northern terminus was at Route 22 (now U.S. Route 54) in southeastern Audrain County. Its southern terminus was in the community of Drake at Route 12 (now U.S. Route 50).
