The original Route 43 was one of the original 1922 Missouri highways. It later became part of Route 13 when that road was extended to the Arkansas state line. The northern terminus of the highway was at Route 16 (now U.S. Highway 60) at Marionville. Its southern terminus was at the Arkansas state line at Blue Eye. Much of the highway is now Route 413.

The highway crossed the James River at Galena on the old Y-Bridge, a bridge which split in two directions midway over the river. That bridge is on the National Register of Historic Places.
