- Sakata Gintoki (坂田銀時 Sakata Gintoki (Seiyū - Tomokazu Sugita)?)
- Sakata Gintoki is the main character of the story. He has naturally permed silver hair, which he blames on his lacking love-life. He rides a motoscooter and wields a bokuto that is from Lake Toya. At first it is believed that he bought it on a class trip because he knocked it over. In reality, the Bokuto he wields is the 'Hoshikudaki', which he bought off of a shopping program and had the maker inscribe "Lake Toya" onto it. His job, as is written on his business card, is a "free-lancer" meaning he'll take any job for nearly any price. He has a sweet tooth and will go into crazed combat if he does not have his sweets (although he does have glycosuria).
- In the past, he was a Joi, someone who fought the Amanto to protect the government. He, Katsura "Zura" Kotaro, Takasugi Shinsuke and Sakamoto Tatsuma are the only mentioned survivors from the Joi. The only reason Gintoki does not join Katsura again to fight the aliens is that he does not like to fight along with "gloomy terrorists". While the reason he decline to join Sakamoto in space travel is that he likes the Earth more than the outer space.
- His name might be a reference to Sakata Kintoki.
- Shimura Shinpachi (志村新八 Shimura Shinpachi (Seiyū - Daisuke Sakaguchi)?)
- Shinpachi works under Gintoki to learn the ways of the samurai. He used to stay at his family's dōjo. However it seems more like he's just trying to help Gintoki pay the rent than to learn. Shinpachi is the captain of the "Otsū-chan's Imperial Guard," a pop-singer's fan club.
- Shinpachi meets Gintoki after Shinpachi was tripped by the ambassador of Chatoransei. Gintoki beats up the ambassador and his guards, not to stand up for Shinpachi but to get revenge for his spilled parfait. Gintoki frames Shinpachi for the crime, which somehow led to him saving Shimura Tae (Otae), Shinpachi's sister, from becoming part of a brothel.
- Despite appearances, Shinpachi is a competent swordsman of his family's Kakidō-Ryu. Which played a large part in saving his sister 2nd time.
- Kagura (神楽 (Seiyū - Rie Kugimiya)?)
- Kagura is from the alien Yato Clan, one of the strongest races in existence. Despite her alien heritage, she appears as a normal, cute human girl in Chinese clothes. She can stop Gintoki's motorscooter single-handedly and has an umbrella that is bullet-proof and can fire bullets from the tip. Her stomach knows no bounds and apparently loves to eat sukonbu. According to a fansite and the OVA as well, she is the first JUMP girl to throw up. [1]
- She originally came to Earth to earn money to help her parents' financial situation. Because of her immense strength, she took to fighting for a gang. After she is ordered to kill a few people, she runs away from the gang is hit by Gintoki's motorscooter. He liberats her, and she threatens him in to letting her work for him. She sleeps in the closet of Gintoki's home.
- She speaks Japanese with a stereotypical Chinese accent (basically adding "-aru" to the ends of her sentences). While she is stronger than most humans, she cannot control her strength perfectly, and is thus happy to have Sadaharu as a pet because she can actually play with him without killing him. She appears to be slightly faster than Okita.
- The Yato clan, according to the gang's boss, is bloodthirsty and has destroyed whole stars. Their weapon of choice appears to be the umbrella.
- Sadaharu (定春 (Seiyū - Mikako Takahashi)?)
- Sadaharu is a giant dog of unknown origins. When sitting on his haunches, he is taller than most humans. He was left in front of Gintoki's apartment in a box that had "To Mr. Freelancer" written on it. Sadaharu chomps on anything smaller than itself, such as Gintoki's and Zura's head. He was named by Kagura, after her first pet Sadaharu, the rabbit whom she killed while having a nightmare. Later it is was revealed that Sadaharu is one of the Guardian Dogs of Japan and was origingally owned by the Witch Sisters.

The main shinsengumi are rivals to Gintoki, Kagura, and Shinpachi. Hijikata is Gintoki's rival, Okita is Kagura's, and Yamazaki is Shinpachi's. Occasionally they are forced to cooperate.
- Kondo Isao (近藤勲 Kondō Isao (Seiyū - Susumu Chiba)?)
- Roughly based on Kondo Isami of the historical Shinsengumi (新選組), the leader of Kyoto's police organization, the Shinsengumi (真選組). He is a bit goofy but nevertheless good-natured and honorable; the members of Shinsengumi are loyal to him. He has a crush on Otae and stalks her, to which Otae responds with merciless beatings. His crush is apparently predicated on the fact that Otae said he was very manly, despite the fact that he's impotent. Other people in manga often call him a gorilla and he refers himself as "Gorilla 13" once because he had been dumped by thirteen girls in one year.
- Hijikata Toshiro (土方十四郎 Hijikata Tōshirō (Seiyū - Kazuya Nakai)?)
- The Vice-Captain of the Shinsengumi. He is very hot-headed and often speaks of seppuku. He is very loyal to Kondo. He loves to fight and sees Gintoki as his rival not only in terms of fighting but also in terms of food (Gintoki loves sweets, unlike Hijikata). He is obsessed with putting mayonnaise on food. Many girls had a crush on him until they discovered his this obsession. He is roughly based on Hijikata Toshizo.
- Okita Sogo (沖田総悟 Okita Sōgo (Seiyū - Kenichi Suzumura)?)
- Okita, roughly based on Okita Sōji, is the best swordsman in the Shinsengumi. Personality-wise, he tends to be light-hearted and somewhat childish, not entirely unlike Kagura. He continuously tries to kill (or at least badly wound) Hijikata whenever he has the chance so that he can become the Vice-Captain. Hijikata once called him "the Prince from the Planet S[adistic]." He bears a grudge against Kagura after an unfinished rock-paper-scissors duel during a sakura-blossom party. He wears a distinctive eye mask when taking a nap during duty. At one point, he refers to himself as the best sniper in the Shinsengumi (not that he is). He often uses a bazooka.
- In volume six Okita's age is given around twenty, but he apparently can get away with drinking.
- Yamazaki Sagaru (山崎退 Yamazaki Sagaru?)
- Yamazaki is a spy, whose specialty is naturally gathering intelligence. Yamazaki is often caught by Hijikata playing badminton by himself, and he is subsequently beaten. His role as a ninja is roughly based on Yamazaki Susumu, but is never really exercised besides one unsuccessful attempt to spy on Gintoki. Despite having a rivalry with Shinpachi, they occasionally see eye-to-eye.

- Terada Ayano/Otose-san (寺田綾乃/お登勢 Terada Ayano/Otose (Seiyū - Kujira)?)
- Gintoki's landlady. He swore to protect her when he ate food offerings to her dead husband. Her real name is Terada Ayano, Otose is a fake "genji" name. She was very pretty when she was young and worked at a restaurant. She secretly fed poor children dumplings for free, and was fired. She is a smoker. Despite constant arguments over Gintoki's general inability to pay his rent, she is confident in his defense of her.
- Katherine (キャサりひ Catherine (Seiyū - Yuu Sugimoto)?)
- An amanto who resembles a cat. She is introduced as a geisha at Otose-san's snack, but reveals herself to be a thief. Despite this, Otose hires her to continue working there. She used to work as a thief with three others vaguely resembling Lupin the Third, Ishizaka Goemon, and Jigen Daisuke. She speaks Japanese with an accent, represented by Katakana characters in the place of Hiragana, but strangely she is the only member of her former crew to speak this way.

- Shimura Tae (志村妙 Shimura Tae (Seiyū - Satsuki Yukino)?)
- Shimura Tae is Shinpachi's older sister. She runs the Kakidōkan Dojo, which was father's failing dojo, with her brother. She is usually addressed as "Otae" (お妙); the "O" is an honorific used to refer to women.
- Despite her sweet face she is one of the most violent characters in Gintama. She mercilessly beats up Kondo Isao and Gintoki. Her weapon of choice seems to be Naginata a Japanese spear. Her cooking is terrible. Her tamagoyaki(i.e. fried egg) is so terrible that Kondo Isao and Gintoki suffer amnesia after eating it. Yamazaki describes is as some sort of weaponry. She has strong principles and believes in maintaining what is precious, even if it means throwing away honor and dignity. She once says that if apologies were enough, seppuku wouldn't exist. Her favourite food is Haagen Dazs branded ice-cream.
- Katsura "Zura" Kotaro (桂小太郎 Katsura Kotarō (Seiyū - Akira Ishida)?)
- Katsura is Gintoki's former comrade as a Ziyo. Even after the Amanto take-over of Japan, Katsura continues to resist the Amanto. His weapon of choice is a metallic grenade. He is hunted by Shinsengumi, the police organization in Kyoto.
- Gintoki often calls him "Zura", to which Katsura indignantly replies "It's not Zura, it's Katsura.". This is due to the reason that "Zura" is means a wig in Japanese. He is sometimes mistaken for a woman. He loves cosplay.
- He is roughly based on one of the three main contributors in Japan's Meiji Restoration, Katsura Kogoro.[citation needed]
- Elizabeth
- Elizabeth, which resembles a giant white duck or a giant penguin, is Katsura's cohort. Although many people consider Elizabeth to be inane and extraneous, Katsura has named the ambiguous avian "Elizabeth" and dotes on it.[citation needed]Elizabeth is something of a painter.
- It's suspected that Elizabeth is actually nothing more than a man wearing a duck costume. In the second version of the ending credits Elizabeth is turned upside-down and his or her white body slips down revealing yellow feet but flesh-colored legs. The online character slideshow shows Elizabeth's beak opening to reveal two glowing eyes.
- Sarutobi Ayame "Sa-chan" (猿飛あやめ Sarutobi Ayame?)
- A woman nick-named Sa-[[chan. She is a ninja who is severely short-sighted. She often loses her glasses to hilarious––or disastrous––effect. She has a crush on Gintoki, but he does not return her love. She is a masochist.
- Takasugi Shinsuke
- Takasugi is the former comrade of the Gintoki and Katsura in the Ziyo. They both studied in the same school when they were kids. Now he is known as the most violent man in Ziyo because has has assassinated many of the Bakufu's main officers. He does not seem to be interested in saving the country like Katsura. Recently he joined the space pirates "Harusame" by offering his former comrades' heads as the ticket to join in.
- His most recognizable accessories are his bandage that covers one of his eyes and his colourful clothes. He is roughly based on the real Meiji Restoration figure Takasugi Shinsaku.[citation needed]
- Sakamoto Tatsuma
- Sakamoto former comrade of Gintoki and Katsura in the Ziyo. He is a good-natured guy with and optimistic to the point of being ridiculous, even in the most dangerous situations. His favourite phase is "HA! HA! HA! It must be XXXXX". He also have a tendency to misspell names––he spelled "Gintoki" as "Kintoki," and Gintoki smashed his face into a control panel.
- Despite of his goofy nature, Sakamoto is actually a successful businessman in the galaxy. He believes that business is a way of protecting his county. He has a private, well-equipped and defensive business fleet. He believes that sacrifices should be made for the good of the community, which is why he choose to abandon his comrades on Earth to travel the space. He is roughly based on the real Meiji Restoration figure Sakamoto Ryoma.[citation needed]
- Hasegawa Taizo
- Formerly worked with the Bakufu. He is fired because Gintoki messed up with the task of looking for the Hata Ouji's pet. Since then, he got fired almost every time in his new job and is constantly seen working various part-time jobs. He and Gintoki are on good terms with one another. He initially believed that humans should focus on appeasing the Amanto, but after his encounter with Gintoki, his mindset changed, which led to him punching Hata out. He often plays pachinko, but rarely ever wins.
- Terakado Otsuu (寺門通 Otsuu (Seiyū - Mikako Takahashi)?)
- An idol pop singer who's song lyrics are often very dirty or controversial. Shinpachi, leader of her "Imperial Guard," first met her when she was a poor singer playing guitar and singing on the streets. Since then, he has helped to save her career many times.
- Prince Hata
- Prince Hata is a squat, purple alien prince who is fond of animals. Much to his dismay, he is often known as "Baka Ouji" (Prince Stupid) due to his simple-mindedness and lack of responsibility with his animal collection. He is a recurring character in Gintama.
- Yagyu Kyuubei
- Firstly introduced at the episode 110 of the manga. She is a female that diguised as a male in order to succeed her family's clan. Her trademark appearance is the eyepatch at her left eye. Because she is raise as a boy, she had a male personality. Although that, she insisted that she want to live as a girl just like her best friend Shimura Tae. She is a master in Yagyu-Ryu sword technique. Her strength is just below his grandfather.
- Her name is roughly based on one of the famous Japanese swordsmanYagyū Jūbei Mitsuyoshi.[citation needed]
- Kawakami Bansai
- He is one of the strongest warrior that works under Takasugi. Little else is known about him except he likes to play music and he is a song writer. He's recognizable accessories are his sunglasses and always wear a pair of headphones.
- His name is roughly based on one of the hitokiri at the Bakumatsu era Kawakami Gensai.[citation needed]
- Okada Nizou
- A blind hitokiri under the employ of Takasugi. His body several modifications with it, including a giant blade on his right arm. He later reveals that his entire upper torso has similar cybernetic modifications. He is eventually defeated by Gintoki with a slash to the face.
- Justaway
- A cylindrical items with crude faces painted on them and fins. Though they seem like harmless merchandise, in actuality, they're bombs being used in a plot against the Bakufu. When Gintoki and Kondo lost their memory, they ended up working at a Justaway factory.
