State Road 101 is a state highway in Duval County, Florida, United States, connecting State Road A1A to the east side of Naval Station Mayport along Mayport Road. Along its route, SR 101 meets the east end of State Road 116. To the south of SR 101, Mayport Road continues as SR A1A to Atlantic Boulevard (State Road 10); SR A1A splits from Mayport Road at SR A1A to reach the Mayport Ferry.

Mayport Road was added to the state highway system in the 1940s as State Road 560. In the 1945 renumbering it was assigned the State Road 101 designation. Later, when SR A1A was realigned from its original northern terminus af Jacksonville to cross the Mayport Ferry, SR 101 was truncated.
