Cortactin is an actin-binding protein of the cytoskeleton. It consists of an N-terminal NTA domain, 6.5 actin binding repeats, a proline-rich region and an SH3 domain. Cortactin is a substrate of both serine/threonine and tyrosine kinases. The protein is thought to couple cellular signalling events (e.g. activation of Src family tyrosine kinases) to rearrangements of the actin cytoskeleton. The gene which encodes cortactin (EMS1) is amplified in several tumors.
