Route 49 is a state route in Connecticut. It was established in 1958, starting in Stonington at Route 2 and ending at Route 14A in Sterling. It goes for 21.74 miles, with a total 18.8 miles of it as a designated state scenic route (in two sections).

The history of Route 49 begins in 1932 when an old Route 49 (now today's Route 272) connected the towns of Torrington and Norfolk from old State Route 312. This route was decomissioned in 1954 as it became part of Route 72. In 1963, it was renamed Route 272.

The current alignment was known as State Highway 216 in the 1920s, and was renumbered in the 1932 renumbering to Route 95. To make way for the new Interstate 95, the route was renumbered again in 1958 to Route 49.
