Brookings County is a county located in the U.S. state of South Dakota. As of 2000, the population was 28,220. Its county seat is Brookings6.

The county was founded July 3, 1871 and was named after Wilmot Wood Brookings, a politician and pioneer of southeastern South Dakota.

According to the U.S. Census Bureau, the county has a total area of 2,084 km² (805 mi²). 2,058 km² (794 mi²) of it is land and 27 km² (10 mi²) of it (1.28%) is water.

As of the census² of 2000, there were 28,220 people, 10,665 households, and 6,217 families residing in the county. The population density was 14/km² (36/mi²). There were 11,576 housing units at an average density of 6/km² (15/mi²). The racial makeup of the county was 96.36% White, 0.31% Black or African American, 0.90% Native American, 1.34% Asian, 0.04% Pacific Islander, 0.30% from other races, and 0.75% from two or more races. 0.88% of the population were Hispanic or Latino of any race.
There were 10,665 households out of which 28.60% had children under the age of 18 living with them, 49.00% were married couples living together, 6.60% had a female householder with no husband present, and 41.70% were non-families. 29.60% of all households were made up of individuals and 8.60% had someone living alone who was 65 years of age or older. The average household size was 2.38 and the average family size was 2.97.
In the county, the population was spread out with 20.80% under the age of 18, 26.80% from 18 to 24, 24.30% from 25 to 44, 17.30% from 45 to 64, and 10.90% who were 65 years of age or older. The median age was 27 years. For every 100 females there were 102.10 males. For every 100 females age 18 and over, there were 101.20 males.
