The B3109 starts in Corsham, at a Give Way junction with the A4. The road then ascends a gentle hill and passes close to the industrial estates at Westwells and the large housing estate at Rudloe, providing an important link to these areas of Corsham. From here, the road passes through gently rolling countryside, predominantly heading downhill, passing Chapel Plaister, then crossing the Five Ways junction with the A365.
The road passes east of South Wraxall, then goes through Bradford Leigh. The road crosses the B3105 at a crossroads just outside Bradford-on-Avon. Through Bradford-on-Avon, the B3109 multiplexes with the A363, passing down the congested Masons Lane then over the Avon Bridge.
Coming out of Bradford-on-Avon, the road climbs a low hill, then is fairly straight and flat from then onwards. The road goes through Wingfield, where there is a traffic lighted junction withe A366 Trowbridge to Radstock road. The road terminates in Rode, 4 miles north east of Frome at a Give Way junction with the A361.
